Food
· Severity: Moderate
· Notes for Consumers: Primaquine may be taken with most foods in order to limit GI side effects, like nausea. However, it is recommended that you do not significantly alter your intake of Grapefruit Juice, or avoid it altogether, when taking Primaquine. Grapefruit juice may increase blood levels of Primaquine by reducing Primaquine metabolism in the body. Too much Primaquine in the blood can lead to side effects such as dizziness, vomiting or low blood counts, which could lead to an increased risk of anemia or infection.
· Notes for Professionals: Overall, the mean bioavailability of primaquine is increased by administration with food; the increase is not thought to be clinically harmful and may contribute to antimalarial efficacy. Administration with food is often recommended to limit GI intolerance.

· Notes for Professionals: Administration of primaquine with grapefruit juice requires caution. The metabolism of primaquine may be reduced when taken with grapefruit juice. In healthy subjects, grapefruit juice increased the mean Cmax of primaquine by 23% and the AUC by 19%; however, there was significant interindividual variability in the results. The authors of the study suggest avoidance of grapefruit juice during primaquine therapy. Alternatively, significant alterations in grapefruit juice intake should be avoided as a precaution during primaquine treatment.
