· Notes for Consumers: Do not take Grapefruit juice or grapefruit-containing foods and Elexacaftor; Tezacaftor; Ivacaftor together without talking to your health care professional. Side effects from Elexacaftor; Tezacaftor; Ivacaftor may get worse if you drink Grapefruit juice or consume grapefruit-containing foods. Notify your prescriber if you have any new or worsening side effects such as dark yellow or brown urine, general ill feeling or flu-like symptoms, light-colored stools, loss of appetite, nausea, right upper belly pain, unusually weak or tired, yellowing of the eyes and skin.

· Notes for Professionals: Advise patients to avoid grapefruit juice or grapefruit-containing foods while taking elexacaftor; tezacaftor; ivacaftor as coadministration may increase elexacaftor; tezacaftor; ivacaftor exposure. Elexacaftor, tezacaftor, and ivacaftor are CYP3A4 substrates (ivacaftor is a sensitive substrate); fosamprenavir is a strong CYP3A4 inhibitor. Coadministration of a strong CYP3A4 inhibitor increased elexacaftor exposure by 2.8- fold, tezacaftor exposure by 4.5-fold, and ivacaftor exposure by 15.6-fold.
