Notes for Consumers: Do not ingest Grapefruit or Grapefruit Juice while on Ivosidenib therapy. Side effects from Ivosidenib may get worse if you take Grapefruit Juice or grapefruit-containing foods. Contact your prescriber immediately if you experience chest pain, dizziness, fainting or falling spells, palpitations, shortness of breath, or a change in your heartbeat (such as a fast or irregular heartbeat).
Notes for Professionals: Advise patients to avoid grapefruit juice while taking ivosidenib due to increased plasma concentrations of ivosidenib, which increases the risk of QT prolongation or other toxicities. Ivosidenib is a CYP3A4 substrate and grapefruit juice is a strong CYP3A4 inhibitor. Coadministration with another strong CYP3A4 inhibitor increased ivosidenib single-dose AUC to 269% of control, with no change in Cmax.
