• Interaction: Grapefruit juice• Severity: Moderate• Notes for Consumers: Do not eat or drink Grapefruit or Grapefruit Juice while taking Polatuzumab Vedotin without talking to your health care provider. Grapefruit Juice or grapefruit-containing foods may increase side effects from Polatuzumab Vedotin.• Notes for Professionals: Advise patients to avoid grapefruit juice while taking polatuzumab vedotin due to the risk of elevated exposure to the cytotoxic component of polatuzumab vedotin, MMAE. MMAE is metabolized by CYP3A4; grapefruit juice is a strong CYP3A4 inhibitor. Strong CYP3A4 inhibitors are predicted to increase the exposure of MMAE by 45%.
