DRONEDARONE (droe NE da rone) treats a fast or irregular heartbeat (arrhythmia). It works by slowing down overactive electric signals in the heart, which stabilizes your heart rhythm. It belongs to a group of medications called antiarrhythmics.

  • Dronedarone 400mg, Oral tablet

    • Interaction: Marijuana
    • Severity: Moderate
    • Notes for Consumers: The effects of marijuana may be increased and side effects may get worse if it is combined with this medicine. Do not drive or operate machinery until you know how this combination will affect you. Contact your health care provider right away if you notice slurred speech, confusion, severe drowsiness, increased heart rate, or any other new or unusual side effects.
    • Notes for Professionals: The incidence of marijuana associated adverse effects may change following coadministration with dronedarone. Dronedarone is an inhibitor of CYP3A4, an isoenzyme partially responsible for the metabolism of marijuana's most psychoactive compound, delta-9-tetrahydrocannabinol (Delta-9-THC). When given concurrently with dronedarone, the amount of Delta-9-THC converted to the active metabolite 11-hydroxy-delta-9-tetrahydrocannabinol (11-OH-THC) may be reduced. These changes in Delta-9-THC and 11-OH-THC plasma concentrations may result in an altered marijuana adverse event profile.

    • Notes for Consumers: Consuming grapefruit juice can increase the side effects of Dronedarone. Contact your prescriber immediately if you experience unexplained muscle aches, muscle pain or tenderness, general weakness or fatigue, side or back pain, or decreased urination while taking Dronedarone.
    • Notes for Professionals: Concomitant use of dronedarone and grapefruit juice should be avoided. Dronedarone is metabolized by CYP3A. Grapefruit juice is a moderate inhibitor of CYP3A4. The concomitant administration of dronedarone and grapefruit juice results in an increase in dronedarone exposure by 3-fold and increase in dronedarone Cmax of 2.5-fold. No data exist regarding the appropriate dose adjustment needed to allow safe administration of dronedarone and grapefruit juice.
