  • Deflazacort 22.75mg/1mL, Oral suspension

    Interaction: Grapefruit juice
    Severity: Major
    Notes for Consumers: Although an interaction is possible, grapefruit juice and deflazacort may be taken together with caution. Side effects may get worse if you take grapefruit juice together with deflazacort. The dose of your Deflazacort will need to be decreased if you also take grapefruit juice. Discuss any new or worsening side effects, like roundness of the face, high blood pressure, extreme irritability or hallucinations, fainting, stomach pain, or water retention, with your health care provider.
    Notes for Professionals: Decrease deflazacort dose to one third of the recommended dosage when coadministered with grapefruit juice. Concurrent use may significantly increase concentrations of 21-desDFZ, the active metabolite of deflazacort, resulting in an increased risk of toxicity. Deflazacort is a CYP3A4 substrate; grapefruit juice is a strong inhibitor of CYP3A4. Administration of deflazacort with clarithromycin, a strong CYP3A4 inhibitor, increased total exposure to 21-desDFZ by about 3-fold.
