ALUMINUM HYDROXIDE; MAGNESIUM CARBONATE (a LOO mi num hye DROX ide; mag NEE zee um KAR bon ate) treats occasional heartburn, indigestion, upset stomach, or other conditions caused by too much stomach acid. It works by reducing the amount of acid in the stomach. It belongs to a group of medications called antacids.

    Notes for Consumers: Do not take Pazopanib with grapefruit, grapefruit juice, or supplements that contain grapefruit extract. Taking Pazopanib with grapefruit or grapefruit juice may increase side effects of Pazopanib. If you accidentally take Pazopanib with grapefruit or grapefruit juice, immediately tell your doctor about any unusual effects.
    Notes for Professionals: Avoid grapefruit juice during pazopanib treatment. Increased pazopanib serum concentrations may occur if grapefruit or grapefruit juice is consumed, possibly resulting in torsade de pointes and/or myelotoxicity. Pazopanib is a substrate for CYP3A4. Grapefruit juice inhibits the cytochrome P-450 3A4 isozyme in the gut wall. Grapefruit juice contains furanocoumarins that are metabolized by CYP3A4 to reactive intermediates. These intermediates form a covalent bond to the active site of the CYP3A4 enzyme, causing irreversible inactivation (mechanism-based inhibition). Consequently, CYP3A4 activity in the gut wall is inhibited until de novo synthesis returns the enzyme to its previous level.
